Weeeell..... I agree with Andrew-- one can never have an absolute favorite. BUT. If backed against a wall and forced to quote a movie, the first thing out of my mouth would either be the on from My Man Godfrey, Bringing Up Baby, or this gem from The Philadelphia Story, delivered by Katharine Hepburn when rather tipsy--
"There was once a Chinaman who drowned trying to kiss the reflection of the moon in the river. He was drunk.

Also, several choice lines from Enchanted April. For instance, the number one best way to stop a conversation heading in the wrong direction:

The best movie to quote EVER (and I think those who have seen it will agreee) is You've Got Mail.
It has a quote for every occasion.

I have a ton of Fave's but that's the only one I could think of!
This may not make sense to some, but there are many excellent quotes from Santa Fe Trail. One of my favorite is when John Brown said, "I have nothing personal against you men, but I will deal harshly with any interference."
The other is when Jefferson Davis asked George Custer (Ronald Reagan), "Wasn't there also a Stuart?" Custer replies (after bragging to a ladyfriend about all of his daring exploits that he singlehandedly performed, "Uh, he was around." You have to see the movie to appreciate it.
